چکیده

BackgroundStreptococcus pneumoniae (S. pneumoniae) of serogroup 19 are mainly represented by serotypes 19A and 19F, which associated with antimicrobial resistance disease. The wzy gene, a component the pneumococcal capsular locus, is target to differentiate 19F PCR-based typing. In last decade, allelic variants wzy19F gene have been described, leading misinterpretation typing results.MethodsA collection 154 serotype S. strains recovered from carriage disease in Brazil was evaluated identify characterize variant isolates.ResultsEleven (7%) isolates were detected identified as belonging ST810 (n = 10) or ST13673 1; single-locus ST810). They mostly diseased patients, susceptible agents tested (except for one multidrug-resistant strain) did not harbor pili genes. Sequences these identical each other those previously described Brazil, but slightly different countries.ConclusionThis study indicated that present geographically driven distribution first uncover phenotypic genetic features lineage occurring since 1989.

Genotypic characterization of Streptococcus pneumoniae serotype 19F in Malaysia.

Streptococcus pneumoniae is an epidemiologically important bacterial pathogen. Recently, we reported the antibiotic susceptibility patterns of a limited collection of pneumococcal isolates in Malaysia with a high prevalence of erythromycin resistant strains. In the present study, 55 of the pneumococcal isolates of serotype 19F were further analysed by pulsed field gel electrophoresis (PFGE) and...

Construction of otherwise isogenic serotype 6B, 7F, 14, and 19F capsular variants of Streptococcus pneumoniae strain TIGR4.

The polysaccharide capsule is the primary virulence factor in Streptococcus pneumoniae. There are at least 90 serotypes of S. pneumoniae, identified based on the immunogenicity of different capsular sugars. The aim of this study was to construct pneumococcal strains that are isogenic except for capsular type. Serotype 4 strain TIGR4 was rendered unencapsulated by recombinational replacement of ...

Streptococcus pneumoniae capsular serotype 19F is more resistant to C3 deposition and less sensitive to opsonophagocytosis than serotype 6B.

The polysaccharide capsule is a major virulence mechanism of Streptococcus pneumoniae, shielding the bacterium from phagocytes. Capsule types may differ in their abilities to resist immune defense. Antibody-mediated complement activation and opsonophagocytosis are crucial in protection against pneumococcus. Conjugate vaccine trials suggest imperfect protection against 19F. We have previously sh...

Purification of capsular polysaccharide produced by Streptococcus pneumoniae serotype 19A.

Streptococcus pneumoniae is a major cause of invasive infection in young infants and older adults. There are currently 90 capsular serotypes identified and 23 serotypes (1, 2, 3, 4, 5, 6B, 7F, 8, 9N, 9V, 10A, 11A, 12F, 14, 15B, 17F, 18C, 19F, 19A, 20, 22F, 23F, and 33F) are responsible for about 90% of invasive disease. Among the more than 90 different S. pneumoniae serotypes, serotype 19A is g...

Characterization of 19A-like 19F pneumococcal isolates from Papua New Guinea and Fiji

Molecular identification of Streptococcus pneumoniae serotype 19F is routinely performed by PCR targeting the wzy gene of the capsular biosynthetic locus. However, 19F isolates with genetic similarity to 19A have been reported in the United States and Brazil. We screened 78 pneumococcal carriage isolates and found six 19F wzy variants that originated from children in Papua New Guinea and Fiji. ...
